Roberts Syndrome
- Roberts Syndrome (Including SC Phocomelia)Autosomal recessive
Caused by loss-of-function changes in both copies of ESCO2, which encodes an enzyme needed for sister chromatid cohesion during cell division. A characteristic finding on chromosome testing is premature separation of the centromeres.
- ClinGen rates the link definitive
- 6 GenCC submitters rate it definitive or strong
▶Actionability
Care involves orthopaedic and limb support, cleft and craniofacial surgery, and developmental assessment. Both parents of an affected child are usually carriers, so each further child has a 1 in 4 chance of being affected; genetic counselling and carrier testing of relatives are available.
How ESCO2 is laid out
- Coding sequence
- Untranslated region
- Intron
- Direction of transcription
- Known disease-causing sites
- Stretches short reads find hard
Exons of the gene's MANE Select transcript (release v1.5), from NCBI and EMBL-EBI, on the GRCh38 reference genome.
Chromosome bands from UCSC's GRCh38 ideogram (2022-10-29).
Nearby genes from Ensembl release 116, on GRCh38.
Exons make up 3.8 kb of the gene's 30.8 kb; 1,806 of those bases code for the protein.

70
positions in ESCO2 where ClinVar classes a change as pathogenic or likely pathogenic
None of them falls in a region short reads find hard.
A whole genome covers all 30.8 kb of ESCO2, introns included, not only the 3.8 kb of exons.
ClinVar pathogenic variants (78)
The ESCO2 variants ClinVar classifies pathogenic or likely pathogenic for any condition, reviewed by at least one submitter with criteria, and the most reviewed of them. It is ClinVar's public record of the gene, never a result.
- Pathogenic 19
- Pathogenic/likely pathogenic 6
- Likely pathogenic 53
