Transcriptome Analysis of TGFBI Knockdown vs Normal Corneal Epithelial Cells: Implications for TGFBI Corneal Dystrophy Treatment
The study used shRNA lentiviral vectors to knock down the TGFBI gene in human corneal epithelial cells and profiled the resulting transcriptome changes. A 70.5% knockdown altered genes linked to inflammation, angiogenesis, scarring and extracellular matrix remodelling, supporting shRNA therapy as a potential treatment for TGFBI corneal dystrophies.

Dante Labs’ involvement
Dante Labs carried out the sequencing for this study. Sequencing was performed on the Illumina HiSeq2500 platform at our facility in Italy, on RNA sequencing libraries from five control human corneal epithelial cell cultures (three negative controls and two scrambled controls) and three shRNA TGFBI knockdown cultures (shRNA1, 2 and 3). The paper reports that these libraries were prepared using the Illumina TruSeq Stranded mRNA library kit according to the manufacturer's instructions, and that RNA extraction was performed by the research team using the Promega ReliaPrep RNA Miniprep System.
