Dante Omics AI, together with Rockefeller University, Giunta Lab, and a network of global partners, has assembled the most complete and accurate diploid reference genome for the RPE-1 cell line – one of the most widely used standards for complex cell engineering experiments.
This milestone provides a true “source code” for researchers worldwide, unlocking deeper insights into structural complexity, genomic stability, and CRISPR-based engineering.
Why this matters
The RPE-1 line is a global reference in cell biology, cancer research, and genome editing. Yet until now, its genome was incomplete, leaving key regions blurry or missing.
Our new assembly changes this:
► Diploid completeness – both maternal and paternal haplotypes resolved across chromosomes.► Ultra-high accuracy – > 99.9999% on multiple chromosomes.► Full coverage of complex regions – centromeres, repeats, and structurally challenging DNA now mapped.► Multi-platform integration – data from Illumina short reads, PacBio HiFi, Oxford Nanopore long reads, and complementary resources.► Open access – assembly RPE1v1.1 is now available through NCBI and UCSC Genome Browser.
